In my setup, membership is owned by an Org, and an Org has a set of employees. The membership is configured such that Employees inherit membership of their employer Orgs.

I created my memberships via Import to Organization. I'm not sure if I missed a step in the import, but currently no employees of these Orgs have inherited memberships.

How should I go about updating or importing Memberships so that this inheritance is created?

I think the inherited memberships for the employees s/b created during Import Memberships (assuming that the Employee -> Employer relationships already exist). Do check that your "Relationship Type" for the Membership Type is 'Employer of' (and not 'Employee of').

You can verify that the basic inheritance is working by doing a test where you manually assign an Employee relationship for an Individual to an Org w/ a membership.

If all this is working, and import is NOT creating the inherited relationships - then it sounds like there's a bug so you should verify that it recreates on the public demo and then file an issue with exact steps to recreate it.

Retested on my end, the problem stemmed from User Error.

I had the inheritance relationship configured incorrectly--it was set to "Employee Of" instead of "Employer Of".
