If we are using civismtp, do we need to run CiviMailProcessor.php in cron?

no you do not need to worry about setting up this script andcron when using CiviSMTP.

CiviSMTP provides a return channel for processing replies, unsibscribes and etc through the soap proxy bundled with your CiviCRM. just make sure that your PHP is compiled with SOAP libraries.
