Error: Failed To Open Stream Line 300

When trying to upgrade CiviCRM from 4.3.3 to 4.3.5, I get now to the Upgrade Now but it also contains this warning-

 Warning: require_once(CRM/Core/ClassLoader.php) [function.require-once]: failed to open stream: No such file or directory in require_once() (line 300 of /home/icuu/www/www/sites/default/civicrm.settings.php).

What do I need to do to fix it?
Do I need to do something before doing the Upgrade Now?

I am a newbie and would be grateful for assistance.

Well, strangely, I elected not to run the upgrade because of the error message until I knew more.
Just tried the upgrade again, but this time there was no error message - and the upgrade seemed to go fine.
So problem sorted.
No assistance need at least not with this issue.
