Today I tried it again, changing the From address for a test mailing to a different email address of mine, and GOT THE REPLY. Previously I had been using my admin email, which I had triple checked for accuracy, but this was the default supplied by CiviMail when "new mailing" was selected, and I had been re-using it in my tests. I suspect CiviMail was not recording a value here unless it was different than the suggested default.
