I've migrated a Drupal site from one server to another, upgrading Drupal to 5.7 and CiviCRM to 2.0 and I ran into this problem.

Any attempt to load a search page for a profile, for example <site root>/civicrm/profile?reset=1&gid=1 gets a 404 'page not found' error.  This was working fine before the migration.  I was getting the same error before the upgrades.

This intranet site depends on searchable staff directories, so any help would be greatly appreciated.

All profiles and groups are configured correctly to allow for searching lists.  Resource urls and civicrm settings all appear to be entered correctly.  Clean urls are not activated

Maybe a change is needed to the server by the webhost?  or changes to a settings config or .htaccess file?  Maybe a mod_rewrite problem?  Please help!

ok so this url works and i'll just go with it:

http://<site root>/index.php?q=civicrm/profile&reset=1&gid=1

The original URL format (<site root>/civicrm/profile?reset=1&gid=1) is correct if CLEAN URLs are enabled for your Drupal site. If they are not enabled - you need to use the format in your second post.
